    How to freeze partner sperm

To support you in freezing partner sperm correctly for use in embryo creation, we have developed this step-by-step guide.

When your patient is having embryo creation treatment using partner sperm, it is essential that the sperm sample is properly frozen before shipment to Cryos. Correct freezing helps preserve sperm quality throughout the process – ultimately giving your patients the best possible chance of a successful outcome.

The sample can be processed using either IUI or the ICI method. Please follow the correct procedure outlined below to ensure optimal results.

Step 1: The sample is analysed by adding 5µL fresh viscous sample to two individual counting chambers and using the CASA motility program.

Step 2: SpermCryo All-round is slowly added directly to the sample in a dropwise manner under constant mixing, to a final dilution of 1:3. Ensure the sample is mixed sufficiently before loading the straws.

Step 3: The sample is loaded onto 0.5 CBS straws, and a small part of the sample is frozen in a separate straw for post-freezing analysis.

Step 4: Straws are frozen using the liquid nitrogen vapour freeze method.

Step 5: A post-freezing analysis is performed on the small post-freezing analysis straw.

Step 6: The straws are moved to long-term storage and kept here until further use

Sperm samples processed using IUI method

Step 1: The sample is analyzed by adding 5µL fresh viscous sample to two individual counting chambers and using CASA motility program.

Step 2: The sample is loaded on to a layered gradient consisting of SpermFilter Ready-to-use gradient 80% and 45%, and centrifuged for 20 min, at 0,3 rcf.

Step 3: After centrifugation the supernatant is removed, and the cell pellet is suspended into 3mL SpermWash medium following 10 min centrifugation at 0.3 rcf.

Step 4: After centrifugation the supernatant is removed. The cell pellet is diluted using SpermWash medium.

Step 5: Freezing medium TEST-yolk buffer with glycerol and gentamicin is added in a dropwise manner, to a final dilution of 1:1.

Step 6: The sample is loaded onto 0.5 CBS straws, and a small part of the sample is frozen in a separated straw, for post freezing analysis.

Step 7: Straws are frozen using the liquid nitrogen vapor freeze method.

Step 8: A post freezing analysis is performed on the small post freezing analysis straw.

Step 9: The straws are moved to long term storage and kept here until further use.

      Testing of partner sperm
Before the sperm sample is provided, the partner must be screened for infectious diseases. When partner sperm is frozen at your clinic, we will require documentation confirming tests for HIV (anti-HIV 1+2), Hepatitis B (HBsAg and anti-HBc), and Hepatitis C (anti-HCV), as required by law.

Shipment of partner sperm to Cryos
Partner sperm must be safely shipped to Cryos prior to treatment. You or your patient can arrange shipping yourself, or you can request Cryos to handle it for an additional fee.
